© 2016, 2010 Elsevier Ltd. All rights reserved. The present chapter sets out to define the usefulness of collagen in corneal tissue engineering. Briefly outlining collagen's role in the normal cornea and its biochemistry, the chapter goes on to describe recent approaches to fabricating collagen-based corneal constructs taking in both top-down and bottom-up rational design, as well as synthetic alternatives to collagen. Particular attention is paid to both decellularization and recellularization techniques, as well as the resulting changes in the collagen gel's ultrastructural, biophysical, and biochemical properties.
