SPM studies of DNA architectures on Au(111) and mica surfaces

Spatially distributed DNA oligomer arrays on Au(111) and DNA networks on mica surfaces were created and investigated. Special attention was paid to investigation of applicability of such structures for conductance measurements through individual DNA molecules. Tapping mode and conductive contact Atomic Force Microscopes (AFM) were used to visualise and analyze the DNA distribution on the surface and to study mechanical and conductive properties of individual molecules. 4 and 10 nm long DNA oligomer arrays on Au(111) were created by self-assembly of monolayers of alkanethiol-conjugated DNA and mercaptohexanol. Networks of plasmid DNA on mica surfaces were created by DNA adsorption from solution. Early stages of network formation were investigated.
