IL-10 gene promoter polymorphism and early-onset periodontal diseases

Abstract

The pathogenesis of early onset periodontal diseases (EOP) is likely to be influenced by complex interactions between host immune responses to specific microbial pathogens and environmental factors. Previous studies indicate that genetic polymorphisms in the interleukin (JL-10 gene may be correlated with modulation of disease expression in inflammatory conditions and may alter levels of IL-10 Production, thus predisposing individuals so disease. We thus sought to determine whether a (CA)- dinucleotide repeat polymorphism in the It-t0 gene Promoter was associated with EOP. An approximately 180 bp fragment of the gene was amplified by PCR using fluorescently labelled primers and genotypes were determined by analysis of fragment sizes by high resolution acrylamide gel electrophoresis, employing ABI Genescan and Genotyper software. We screened DNA samples of 72 EOP patients and 73 controls with no history of EOP. The allele frequencies in the patient group were 3 (2%), 3 (2%), 69 (4g%/), 5 (3%), 7 (5%), 15 (10%), 32 (22%/), g (6%/), assay was then 2 (1'%) and 0 (OYYs6) and is the control group 4 (3%/), 5 (3%), 61 (41%/), 13 (90/6), 13 (9h/), 14 (10h/), 26 (lg%), g (5%/), 1(1%/) and 1 (1%) for alleles containing 19 to 28 repeats units respectively. Statistical analysis applying "CLUMP" software (Sham & Curtis, Ann Hum Genet 59:97-105, 1995) revealed no significant difference in allele frequencies or genotype distribution between the two populations (p=0.273 and p=0.597 respectively). We conclude that the methodology described is useful for the analysis of repeat unit polymorphisms, however, our study provides no evidence for an association between the (CA)2 dinucleotide repeat polymorphism in the IL-10 promoter region and susceptibility to EOP.
