Assessing MicroRNA Profiles from Low Concentration Extracellular Vesicle RNA Utilizing NanoString nCounter Technology

Abstract

The Author(s), under exclusive license to Springer Science+Business Media, LLC, part of Springer Nature.Extracellular vesicles (EV) are rich in small RNA; however, a frequent caveat can be low abundance of EV RNA content, especially in clinical studies. NanoString MicroRNA Assays allow for multiplexed profiling of n = 800 mature microRNAs and can be applied to assess EV microRNA cargo. Here, we describe a method to adapt NanoString nCounter microRNA profiling to assess mature microRNA expression in low-concentration RNA samples, including concentrating the RNA, quantifying the RNA, and performing the NanoString protocol. Twelve samples can be assessed at one time using this method.
