Abstract

Purpose: Over 90 independent genome-wide association signals have been reported for osteoarthritis (OA). The vast majority of these risk alleles are thought to mediate their effect through altered expression of a target gene, and are known as expression quantitative trait loci (eQTL). To elucidate the mechanism of risk and target genes at these loci, detailed molecular characterisation of the signals is necessary. Epigenetic mechanisms within chondrocytes can also alter gene expression and underlie regulation of gene expression by single nucleotide polymorphisms (SNPs).
